That's a pretty good sample routine here which provides a good guideline to how much to exercise each body part. I've linked to the page it's from at the bottom of the post.

The exercises that most of the big guys swear by are:

squat
deadlift
military/shoulder press
(incline) bench press
chins
dips

Concentrate on compound exercises - those are the ones that exercise a wide range of muscles rather than targeting one specific one (on exrx.net they're the exercises listed in bold). Most people advise to put the big, compound exercises at the beginning of your workout, with any isolation exercises at the end. But play around with it, see what feels good.

And remember to warm up beforehand - 5 mins cardio beforehand, and a warmup set at about half-weight before your real sets.
